Objective: To establish a method for the content determination of ellagic acid and piperine in pomegranate lotus powder, a Tibetan medicine, in order to improve its quality standard. Methods: The content of ellagic acid in pomegranates and chebula was determined by High Performance Liquid Chromatography (HPLC). The column was ODS-2 HYPERSIL C18 (4.6 mm×250 mm, 5 μm), the mobile phase was acetonitrile-0.2% phosphoric acid (15:85), the detection wavelength was 254 nm, the flow rate was 1.0 mL·min-1, and the column temperature was 30℃. HPLC was used to determine piperine of long pepper in the preparation. The column was ODS-2 HYPERSIL C18 (4.6 mm×250 mm, 5 μm), the mobile phase was acetonitrilewater (40:60), the flow rate was 1.0 mL·min-1, the detection wavelength was 343 nm, and the column temp erature was 30℃. Results: There was a good linear relationship (r=0.9999) between 5.065 and 25.325 μg for ellagic acid, and the average recovery was 97.5% with RSD of 1.5% (n=6). There was a good linear relationship (r=1.0000) between 10.414 and 52.071 μg for piperine, with an average recovery of 99.6% and RSD of 0.6% (n=6). Conclusion: This method could accurately determine the content of ellagic acid and piperine in pomegranate lotus powder, which was helpful to improve the quality standard of pomegranate lotus powder.
