摘要

目的:研究苗药黑龙骨茎叶对胶原诱导性关节炎(Collagen-induced Arthritis,CIA)模型大鼠类风湿性关节炎(Rheumatoid Arthritis,RA)作用,探究黑龙骨茎叶抗RA作用的潜在开发利用度。方法:SD大鼠随机选取8只作为正常组,其余大鼠建立CIA模型。造模成功后随机分为模型组,黑龙骨茎叶高剂量组(100 mg·kg-1)、低剂量组(25 mg·kg-1),雷公藤多苷(TG)组(10 mg·kg-1),每组8只。在试验因素干预后,对各组大鼠AI评分、足趾厚度及病理形态变化进行分析比较。结果:给药前后,正常组与模型组比较有极显著差异(P<0.01),说明造模成功。给药后,大鼠左、右足趾厚度测量值显示,模型组与黑龙骨茎叶高、低剂量组相比均有显著性差异(P<0.05);AI评分显示,黑龙骨茎叶高、低剂量组与模型组相比,存在显著差异,组内比较中,黑龙骨茎叶低剂量组的消肿效果优于高剂量组(P<0.05)。病理切片结果显示,与正常组相比,模型组大鼠滑膜组织内见大量、弥漫性炎性细胞浸润;与模型组相比,黑龙骨茎叶低剂量组大鼠滑膜组织内炎性细胞浸润减少。结论:黑龙骨茎叶表现出了良好的抗RA效果,具有潜在的抗RA开发利用价值。

Abstract

Objective: To investigate the eff ects of Periploca forrestii Schltr. stem leaves on collagen-inducedarthritis (CIA) rats model of rheumatoid arthritis (RA), and to explore the potential exploitation of the anti-RA effect of Periploca forrestii Schltr. stem leaves. Methods: Eight SD rats were randomly selected as thenormal group, and the remaining rats were established as CIA models. After successful modeling, the rats wererandomly divided into model groups, Periploca forrestii Schltr. stem leaves high-dose (100 mg·kg-1) and lowdose(25 mg·kg-1) groups, tripterygium glycosides (TG)(10 mg·kg-1) group, with 8 rats in each group. After theintervention of experimental factors, the AI scores, toe thickness and pathological changes of rats in each groupwere analyzed and compared. Results: Before and after administration, there was a highly signifi cant diff erence(P<0.01) between the normal group and the model group, indicating successful modeling. After administration,measurements of the thickness of the right and left toes of rats showed signifi cant diff erences (P<0.05) between the model group and the high and low dose groups of Periploca forrestii Schltr. stem leaves. AI scores showedthat there was a signifi cant diff erence between the high and low dose groups of Periploca forrestii Schltr. stemleaves compared with the model group, and in the intra-group comparison, the swelling reduction eff ect of thelow dose group of Periploca forrestii Schltr. stem leaves was better than that of the high dose group (P<0.05).The pathological section results showed that a large number of diffuse inflammatory cells infiltrated in thesynovial tissue of rats in the model group compared with the normal group. Compared with the model group, theinfl ammatory cells infi ltrated in the synovial tissue of rats in the low-dose group of Periploca forrestii Schltr. stemleaves were reduced. Conclusion: The stem leaves of Periploca forrestii Schltr. have shown good anti-RA eff ectsand have potential value for anti-RA development and utilization.
